Overview
Baricitinib is a small molecule Janus kinase (JAK) inhibitor, specifically targeting JAK1 and JAK2. It is used as an immunomodulatory agent to treat moderate to severe rheumatoid arthritis in adults who have had an inadequate response to one or more tumor necrosis factor (TNF) antagonists. Baricitinib is also approved for the treatment of severe alopecia areata and for hospitalized patients with COVID-19 requiring supplemental oxygen or mechanical ventilation. By inhibiting JAK1 and JAK2, baricitinib disrupts pro-inflammatory cytokine signaling pathways, thereby reducing inflammation and immune-mediated tissue damage[1][5][7].
