This is a combination of three Janus kinase (JAK) inhibitors: baricitinib, tofacitinib, and upadacitinib. Each is an oral small molecule drug that inhibits one or more members of the JAK family of enzymes (JAK1, JAK2, JAK3, TYK2), which are involved in cytokine signaling pathways that mediate immune and inflammatory responses. These drugs are primarily used for autoimmune diseases such as rheumatoid arthritis and other inflammatory conditions. Baricitinib selectively inhibits JAK1 and JAK2; tofacitinib mainly targets JAK1 and JAK3; upadacitinib is a selective inhibitor of JAK1[6][9]. The combination would theoretically provide broad inhibition across multiple cytokine pathways but may increase the risk for adverse effects such as infections or blood clots.
