BBI-2000

BBI-2000 is a topical "soft" Janus kinase (JAK) inhibitor that was being developed by Fresh Tracks Therapeutics (formerly Brickell Biotech) for the treatment of inflammatory skin conditions. As a soft drug, BBI-2000 is designed to be metabolically unstable; it is active when applied topically to the skin but is rapidly degraded into an inactive metabolite upon entering the systemic circulation. This mechanism is intended to provide the anti-inflammatory benefits of pan-JAK inhibition locally while avoiding the systemic safety concerns (such as cytopenias or thromboembolism) typically associated with oral JAK inhibitors. The compound completed a Phase 1 study for contact hypersensitivity before the developer announced plans for liquidation and ceased further development of its pipeline in late 2023.
