This biparatopic antibody-drug conjugate (ADC) targets Delta-like ligand 3 (DLL3) and is being developed by Earendil Labs for the treatment of small cell lung cancer (SCLC) and other neuroendocrine neoplasms. Engineered using an AI-guided antibody development platform, the drug features a biparatopic antibody designed to bind two distinct, non-overlapping epitopes on the DLL3 protein. This design is intended to enhance internalization efficiency and subsequent delivery of the cytotoxic payload compared to traditional monospecific antibodies. DLL3 is a Notch pathway ligand that is highly expressed on the surface of neuroendocrine tumor cells but has minimal expression in healthy tissues, making it a selective target for ADC therapy. Preclinical studies presented at AACR 2026 demonstrated that this ADC exhibits potent cytotoxicity across various DLL3-expressing cell lines and significant anti-tumor activity in cell-derived xenograft (CDX) models, with a favorable safety profile supported by studies in transgenic mice and non-human primates.
