**CC-90011 + cisplatin + etoposide + nivolumab** is a multi-agent investigational combination regimen studied primarily for the treatment of extensive stage small cell lung cancer (ES-SCLC). The regimen combines: - **CC-90011**, an oral, potent, selective, and reversible inhibitor of lysine-specific demethylase 1 (LSD1/KDM1A), which acts as an epigenetic modulator and shows antitumor activity in neuroendocrine malignancies by reactivating tumor-suppressing genes and decreasing tumor-promoting genes. - **Cisplatin**, a platinum-based chemotherapeutic agent that crosslinks DNA, leading to DNA damage and apoptosis in cancer cells. - **Etoposide**, a topoisomerase II inhibitor that disrupts DNA replication and transcription, causing DNA strand breaks and apoptosis. - **Nivolumab**, a monoclonal antibody immune checkpoint inhibitor targeting programmed death-1 (PD-1), thereby enhancing T-cell mediated antitumor immunity. This combination was studied in Phase 1b in ES-SCLC to investigate safety, dose finding, and preliminary efficacy, and a related combination (CC-90011 + nivolumab) continues in Phase 2 trials.[2][6][4]
