CD19 CAR mRNA-tLNP is an investigational in vivo CAR-T cell therapy platform that utilizes targeted lipid nanoparticles (tLNPs) to deliver mRNA encoding a chimeric antigen receptor (CAR) directly to T cells within the body. The lead candidate, CPTX2309 (developed by Capstan Therapeutics and acquired by AbbVie), employs a CD8-targeted LNP containing a novel ionizable lipid and a CD8-targeting antibody to selectively transfect CD8+ cytotoxic T cells. This process transiently engineers the T cells to express a fully human anti-CD19 CAR, which then targets and eliminates pathogenic CD19+ B cells. This approach is designed to treat B cell-mediated autoimmune disorders and B cell malignancies without the need for complex ex vivo manufacturing or viral vectors. Research also explores different mRNA formats, such as circular mRNA, to enhance the durability and level of CAR expression compared to linear counterparts.
