CDH17 nanoCAR circular RNA with CD11b nanobody-targeted lipid nanoparticles is an experimental in situ cell therapy candidate being developed by Citil Pharma for the treatment of colorectal cancer. The therapeutic consists of circular RNA (circRNA) encoding a nanobody-based chimeric antigen receptor (nanoCAR) specific for Cadherin-17 (CDH17), a membrane protein highly expressed in digestive tract tumors. This circRNA is encapsulated within lipid nanoparticles (LNPs) that are surface-decorated with CD11b-targeting nanobodies. These targeted LNPs selectively deliver the circRNA to NK and NKT cells in vivo, enabling the generation of CAR-expressing effector cells directly within the patient. This approach aims to bypass the logistical complexities and high costs of traditional ex vivo CAR cell manufacturing while providing a dual-acting immune response against solid tumors.
