CEACAM5-targeted clickable antibody with MMAE protodrug is a preclinical oncology therapeutic developed by Shasqi, Inc. using its proprietary Click Activated Protodrugs Against Cancer (CAPAC®) technology. This system is a first-in-class pretargeting approach consisting of two components: a clickable antibody binder directed against Carcinoembryonic Antigen-Related Cell Adhesion Molecule 5 (CEACAM5) and a chemically attenuated, clickable protodrug of the potent cytotoxin monomethyl auristatin E (MMAE). The strategy involves sequential administration where the binder first localizes to the tumor; after systemic clearance of the binder, the protodrug is administered and undergoes a bioorthogonal in vivo click chemistry reaction with the binder at the tumor site. This reaction restores the cytotoxic activity of MMAE locally, thereby maximizing anti-tumor efficacy while minimizing systemic off-target toxicities typically associated with conventional antibody-drug conjugates (ADCs). Preclinical studies have demonstrated robust tumor control and regression in CEACAM5-positive xenograft models of pancreatic, lung, and gastric cancers.
