Overview

CJ-15314 is a novel, highly selective small molecule inhibitor of Janus kinase 1 (JAK1), developed for the treatment of autoimmune and inflammatory diseases such as rheumatoid arthritis, atopic dermatitis, and psoriasis. It demonstrates robust efficacy in preclinical models of rheumatoid arthritis and exhibits a differentiated safety profile compared to other JAK inhibitors. Mechanistically, CJ-15314 inhibits JAK1 with high selectivity—showing approximately 18-fold greater selectivity over JAK2 and 83-fold over JAK3 in biochemical assays. In human whole blood assays, it is significantly more potent against IL6-induced pSTAT1 inhibition via JAK1 than against GM-CSF-induced pSTAT5 inhibition via JAK2. The drug is being developed by HK inno.N (originator: CJ HealthCare) and is administered orally as capsules or topically as ointment[1][2][3][4][5][6].
