Overview

CP-SOCS3 (also known as MTM-SOCS3) is a first-generation cell-permeable recombinant protein therapy developed by Cellivery Therapeutics. It consists of the Suppressor of Cytokine Signaling 3 (SOCS3) protein fused to a membrane translocating motif (MTM), a hydrophobic cell-penetrating peptide derived from the signal peptide of fibroblast growth factor 4 (FGF4). SOCS3 is a negative regulator of the JAK/STAT signaling pathway, which is frequently overactivated in various cancers. By delivering functional SOCS3 protein directly into cells, CP-SOCS3 aims to inhibit the phosphorylation of STAT1 and STAT3, thereby suppressing tumor cell proliferation and migration. While it demonstrated proof-of-concept in preclinical models, it was characterized by low solubility and yield, leading to the development of the improved successor, iCP-SOCS3.
