Decernotinib is an orally administered small molecule inhibitor that selectively targets Janus kinase 3 (JAK3). It was developed by Vertex Pharmaceuticals for the treatment of rheumatoid arthritis and other autoimmune disorders. Decernotinib demonstrates approximately five-fold selectivity for JAK3 over other JAK family members (JAK1, JAK2, TYK2), aiming to modulate immune activity by inhibiting cytokine signaling through receptors that utilize the common gamma chain (such as IL-2, IL-4, IL-7, IL-9, IL-15 and IL-21). Clinical trials showed efficacy in improving symptoms of rheumatoid arthritis; however, development was discontinued before regulatory approval[1][2][5][6][7].
