Deuruxolitinib is an orally administered, small molecule, deuterated Janus kinase inhibitor, targeting JAK1 and JAK2 selectively. It is indicated for the treatment of adults with severe alopecia areata, an autoimmune-mediated disease that results in unpredictable hair loss due to immune attack on hair follicles. The deuterated modification increases its pharmacologic duration compared to non-deuterated analogs. Deuruxolitinib disrupts the JAK-STAT pathway, reducing cytokine-mediated inflammatory responses involved in the pathogenesis of alopecia areata. The drug was developed and is manufactured by Sun Pharma, sold under the brand name Leqselvi. FDA approval for severe alopecia areata was granted in July 2024[1][2][4][5][8].
