The conjugate binds and neutralizes soluble tumor necrosis factor alpha at inflamed tissue and uses a neutrophil elastase-cleavable linker to locally release the Janus kinase inhibitor tofacitinib, aiming to combine TNF alpha blockade with inflammation-site-restricted JAK inhibition while reducing systemic exposure to free tofacitinib.
