ExoCBD is an exosome-encapsulated formulation of cannabidiol (CBD) developed for the targeted oral delivery and treatment of breast cancer, particularly triple-negative breast cancer (TNBC). This delivery system utilizes exosomes, specifically those derived from bovine colostrum, to overcome the poor oral bioavailability and gastrointestinal instability of free CBD. By encapsulating CBD, the formulation protects the drug from acidic degradation in the stomach and facilitates time-dependent release in the intestines. Preclinical studies have demonstrated that ExoCBD (and its folic acid-functionalized variant, FA-ExoCBD) enhances tumor targeting and retention, leading to significant inhibition of orthotopic tumor growth. Mechanistically, the formulation modulates the expression of over 1,000 genes, including the downregulation of pro-metastatic markers like IL13RA2 and tumor biomarkers such as TRPM2 and SAMHD1, while upregulating tumor suppressors like PRDM1.
