FMX114 is a proprietary investigational topical combination gel developed by Vyne Therapeutics for the treatment of mild-to-moderate atopic dermatitis. It combines two small molecule drugs— tofacitinib, a Janus kinase (JAK) inhibitor, and fingolimod, a sphingosine 1-phosphate receptor modulator. Tofacitinib acts intracellularly to reduce inflammation by inhibiting cytokine release from inflammatory cells via JAK1 and JAK2 inhibition. Fingolimod acts extracellularly as an S1P1 modulator to inhibit migration of inflammatory cells and may also support skin barrier recovery by upregulating filaggrin expression. The formulation is designed to concentrate anti-inflammatory effects in the skin while minimizing systemic exposure. FMX114 has been evaluated in Phase 1b/2a clinical trials for atopic dermatitis but did not meet its primary efficacy endpoints in Phase 2a[1][4][5][6][7].
