Follistatin mRNA-loaded lipid nanoparticles are an experimental mRNA-based therapeutic designed to treat metastatic ovarian cancer and its associated cachexia. The therapy utilizes lipid nanoparticles (LNPs) to deliver mRNA encoding follistatin (FST), a natural antagonist of Activin A (ActA). Activin A is often overexpressed in gynecological and other cancers, where it promotes tumor growth, metastasis, and muscle atrophy. By inducing transient expression of FST, the LNPs inhibit ActA activity, leading to reduced tumor progression, decreased malignant ascites, and reversal of muscle wasting. Preclinical studies in murine models conducted by researchers at Oregon Health & Science University have shown that this therapy, particularly when combined with cisplatin, significantly extends survival and improves muscle mass.
