Overview

Frevecitinib is a first-in-class inhaled pan-Janus kinase (JAK) inhibitor in clinical development for the treatment of asthma that remains inadequately controlled despite standard-of-care inhaled maintenance therapies. It is specifically formulated as a dry powder for targeted lung delivery via single-capsule inhaler, aiming to achieve therapeutic concentrations in the lungs while minimizing systemic exposure. Frevecitinib inhibits JAK1, JAK2, JAK3, and TYK2 enzymes involved in pro-inflammatory signaling pathways central to asthma pathophysiology. In Phase 1b studies, it demonstrated clinically relevant reductions in fractional exhaled nitric oxide (FeNO), a key marker of airway inflammation, across patients with mild to moderate-to-severe asthma—including those less responsive to other therapies due to low blood eosinophil counts. The drug is being developed by Kinaset Therapeutics and has received FDA clearance for investigational new drug (IND) application; a Phase 2b dose-ranging trial is planned for mid-2025[8][10].
