GDC-0214 is a potent, selective small molecule inhibitor of Janus kinase 1 (JAK1) developed for inhaled administration as a potential treatment for asthma. The drug was designed to achieve targeted JAK1 inhibition in the lungs while minimizing systemic exposure and associated toxicity. By inhibiting JAK1, GDC-0214 blocks signaling pathways mediated by cytokines such as interleukin (IL)-4 and IL-13 that are implicated in the pathogenesis of asthma. Clinical studies demonstrated dose-dependent reductions in fractional exhaled nitric oxide (FeNO), a biomarker of airway inflammation dependent on JAK1-mediated pathways. The drug was developed by Genentech (a subsidiary of Roche) but its development for asthma has been discontinued after phase I trials.
