glucose-dependent insulinotropic polypeptide + insulin refers to a combination therapy involving the incretin hormone glucose-dependent insulinotropic polypeptide (GIP) and insulin. GIP is a 42-amino acid peptide hormone secreted by intestinal K-cells that enhances insulin secretion from pancreatic beta cells in a glucose-dependent manner and also influences lipid metabolism and bone density. Insulin is a peptide hormone essential for regulating blood glucose levels by promoting glucose uptake in peripheral tissues and inhibiting hepatic glucose production. This combination is primarily investigated for the management of diabetes mellitus, where GIP or GIP receptor agonists are used as adjuncts to exogenous insulin therapy to improve glycemic control, potentially reduce the required insulin dose, and mitigate weight gain. Clinical research, such as retrospective studies conducted by Shanghai General Hospital, has evaluated the real-world impact of adding glucose-lowering agents that affect the GIP pathway (such as DPP-4 inhibitors, which increase endogenous GIP levels) to standard insulin regimens in patients with type 1 diabetes.
