Golixitinib is a potent and highly selective Janus kinase 1 (JAK1) inhibitor developed for the treatment of T-cell malignancies. Originally discovered by AstraZeneca (as AZD4205) and subsequently licensed to Dizal Pharma (as DZD4205), the drug targets the JAK-STAT signaling pathway, which is frequently activated in T-cell lymphomas. In June 2024, golixitinib received its first global approval from China's National Medical Products Administration (NMPA) for the treatment of relapsed or refractory peripheral T-cell lymphoma (PTCL) in adult patients who have failed at least one prior systemic therapy. The drug is administered orally and is being evaluated in various clinical trials, both as a monotherapy and in combination with other agents like the HDAC inhibitor chidamide, particularly for cutaneous T-cell lymphoma (CTCL).
