iCP-SOCS3 is an intracellular protein therapeutic that utilizes advanced macromolecule transduction domains (aMTDs) to enable its delivery into cells. It is designed to selectively target tumors that are dependent on JAK/STAT signaling. The drug functions by delivering the Suppressor of Cytokine Signaling 3 (SOCS3) protein, which acts as a feedback inhibitor of the JAK/STAT pathway. Its mechanism involves directly inhibiting the catalytic activity of Janus kinases (JAK1, JAK2, and TYK2) and promoting the proteasomal degradation of both JAKs and cytokine receptors (such as gp130, G-CSF, and leptin receptors) through the recruitment of E3 ligase components. This dual action leads to the suppression of cytokine-induced signaling, thereby inhibiting cancer cell proliferation and migration, and suppressing tumor growth in xenograft models.
