Overview

IGEM-007 is a first-in-class monoclonal antibody of the IgE isotype targeting the High Molecular Weight Melanoma Associated Antigen (HMW-MAA), also known as Chondroitin Sulfate Proteoglycan 4 (CSPG4). Developed by IGEM Therapeutics, it is designed to treat malignant melanoma by leveraging the unique properties of the IgE immune pathway. Unlike conventional IgG antibodies, IgE has a significantly higher affinity for its receptors (FcεRI) on effector cells such as monocytes, macrophages, and eosinophils, and it naturally resides in tissues rather than blood. This allows for potent antibody-dependent cell-mediated cytotoxicity (ADCC) and phagocytosis (ADCP) against solid tumors. IGEM-007 is the first IgE antibody to enter clinical development for melanoma, aiming to overcome the limitations of standard IgG-based immunotherapies in the tumor microenvironment.
