Overview

INCB047986 is an orally bioavailable small molecule inhibitor of Janus-associated kinases (JAK), specifically targeting Janus kinase 1 (JAK1) and Janus kinase 2 (JAK2). It was developed by Incyte for its potential antineoplastic and immunomodulatory activities. By inhibiting JAK1 and JAK2, INCB047986 disrupts the JAK/STAT signaling pathway, which is involved in cytokine-mediated processes such as cell proliferation, survival, and immune regulation. This mechanism makes it a candidate for treating various cancers—including advanced or metastatic pancreatic cancer, breast cancer (triple-negative), urothelial cancer—as well as autoimmune diseases like rheumatoid arthritis and myelodysplastic syndrome resistant to erythropoiesis-stimulating agents[1][2][3][6][7]. Clinical development included phase 1 studies in oncology and trials in rheumatoid arthritis; however, global development status has been discontinued[4].
