Overview

This is a combination of two endogenous pro-inflammatory cytokines—interleukin-1β (IL-1β) and tumor necrosis factor alpha (TNF-α). Both are key mediators in the inflammatory response and play central roles in immune regulation, cell signaling, and pathophysiology of various diseases. When administered together or co-expressed at high levels, they exhibit synergistic effects that can amplify inflammation or tissue injury. In experimental settings, this combination has been shown to induce myocardial depression at lower concentrations than either cytokine alone[1], promote regression of endothelial capillary tubes[5], upregulate inflammatory enzymes such as phospholipase A2[2], and increase expression of adhesion molecules like ICAM-1 and VCAM-1 on vascular cells[3]. Their combined activity is implicated in the pathology of conditions such as sepsis-associated cardiac dysfunction[1] and neuroinflammation relevant to neurodegenerative diseases like Parkinson’s disease[4]. This combination is not an approved pharmaceutical product but may be used experimentally to model or study inflammatory processes.
