INV441 is an autologous cell therapy developed by invIOs that utilizes the EPiC platform to treat glioblastoma. The therapy involves isolating tumor-infiltrating lymphocytes (TILs) from the patient, modifying them ex vivo using siRNA to silence Cbl-b expression (an intracellular immune checkpoint), and then administering these modified TILs directly into the tumor. By inhibiting Casitas B-lineage lymphoma proto-oncogene B (Cbl-b), a negative regulator of T cell activation, INV441 aims to enhance local anti-tumor immune responses and promote tumor cell killing. This approach has demonstrated strong preclinical proof of concept in murine glioma models and is designed for rapid out-patient manufacturing with short vein-to-vein times. A first-in-human Phase 1 study in glioblastoma is planned for 2025 in collaboration with Dana-Farber Cancer Institute[1][3][7][8].
