Overview

Izencitinib is an orally administered, gut-selective small molecule pan-Janus kinase (JAK) inhibitor developed for the treatment of inflammatory bowel diseases such as ulcerative colitis and Crohn's disease. It binds with high affinity to JAK family members JAK1, JAK2, and JAK3 and was designed to maximize efficacy in gastrointestinal tissues while minimizing systemic exposure and associated adverse effects typical of systemic JAK inhibition. The drug was developed by Theravance Biopharma with the aim of targeting inflammation locally in the GI tract to reduce risks like immunosuppression or malignancy seen with other non-selective or systemically absorbed JAK inhibitors[1][7][8].
