Jaktinib is a novel small molecule inhibitor targeting Janus kinases (JAK1, JAK2, JAK3, TYK2) and activin A receptor type 1 (ACVR1). It blocks the JAK-STAT signaling pathway by inhibiting these kinases and thereby modulates immune responses and cytokine signaling. Additionally, inhibition of ACVR1 reduces hepcidin transcription to improve iron metabolism and anemia in myelofibrosis patients. Developed as a first-in-class drug in China for intermediate- or high-risk myelofibrosis (MF), it has also entered clinical trials for other indications such as alopecia areata, atopic dermatitis, psoriasis, and ankylosing spondylitis[2][3][4][6].
