Jaktinib + glucocorticoids is a combination therapy being evaluated as a first-line treatment for grade II-IV acute graft-versus-host disease (aGVHD). The regimen consists of jaktinib (also known as gecacitinib), a small molecule pan-Janus kinase (JAK) inhibitor, and glucocorticoids (typically methylprednisolone). Jaktinib works by inhibiting the JAK-STAT signaling pathway, which is central to the production and action of pro-inflammatory cytokines that drive the immune-mediated damage in aGVHD. Glucocorticoids provide complementary immunosuppression by binding to the glucocorticoid receptor and modulating gene expression to reduce inflammation. This combination is currently being studied in a Phase 1/2 clinical trial led by the First Affiliated Hospital of Zhejiang University to determine the recommended Phase 2 dose and assess preliminary efficacy and safety in patients following allogeneic hematopoietic stem cell transplantation.
