LNP-circRNAE7E6 is a circular RNA (circRNA) therapeutic vaccine encapsulated in lipid nanoparticles (LNPs), developed by Therorna for the treatment of HPV16-associated malignancies, such as cervical and head and neck cancers. The vaccine encodes a fused, non-oncogenic HPV16 E7/E6 antigen designed with endoplasmic reticulum (ER)-targeting signals and helper epitopes to optimize antigen processing and T-cell activation. By leveraging the stability and high translation efficiency of the circRNA platform, LNP-circRNAE7E6 aims to elicit robust and durable anti-tumor CD8+ T-cell responses. Preclinical studies have demonstrated its ability to reshape the tumor microenvironment, induce complete tumor regression in TC-1 tumor-bearing mice, and sensitize tumors to PD-L1 blockade, leading to improved survival outcomes.
