LW402 is a next-generation, orally administered small molecule inhibitor that selectively targets Janus kinase 1 (JAK1). It was developed to address the need for more selective JAK inhibition in autoimmune diseases, aiming to improve safety and efficacy compared to earlier broad-spectrum JAK inhibitors. Preclinical studies demonstrated potent nanomolar activity against JAK1 with approximately 45-fold selectivity over JAK2-dependent signaling. LW402 has shown significant efficacy in animal models of rheumatoid arthritis, reducing inflammation and protecting joint tissue. Clinical development has focused on rheumatoid arthritis and other autoimmune conditions such as atopic dermatitis and vitiligo[1][2][8][9].
