Overview

Momelotinib + capecitabine is an investigational **combination therapy** being evaluated for the treatment of **relapsed/refractory metastatic pancreatic ductal adenocarcinoma**. **Momelotinib** is a small molecule **dual inhibitor of Janus kinase 1 (JAK1) and Janus kinase 2 (JAK2)**, as well as an antagonist of **Activin receptor type 1 (ACVR1)**; it exerts effects on the hematological and fibrotic aspects of disease and modulates iron metabolism. **Capecitabine** is an oral **antimetabolite prodrug** of 5-fluorouracil (5-FU), used extensively in the treatment of various cancers, including pancreatic cancer, and acts as a **thymidylate synthase inhibitor** interfering with DNA synthesis in rapidly dividing cells. The combination is being studied with oxaliplatin for safety, tolerability, and maximum tolerated dose in pancreatic ductal adenocarcinoma.
