NCTX-01 is a first-in-class, dual-targeting *in vivo* CAR-T therapy developed by NanoCell Therapeutics for B-cell malignancies, including diffuse large B-cell lymphoma (DLBCL) and follicular lymphoma. It utilizes a proprietary targeted lipid nanoparticle (tLNP) platform (NCtx) to deliver minicircle DNA encoding a dual-CAR construct (targeting CD19 and CD22) and SB100x transposase mRNA directly to T cells within the patient's body. The nanoparticles are functionalized with anti-CD7 and anti-CD3 binders to ensure specific delivery to T cells. This non-viral, DNA-based approach aims to eliminate the need for complex *ex vivo* cell manufacturing, potentially offering a more accessible and scalable alternative to traditional CAR-T therapies.
