Nezulcitinib is a lung-selective, inhaled small molecule inhibitor of the Janus kinase (JAK) family, developed for its immunomodulatory and anti-inflammatory properties. It acts as a pan-JAK inhibitor, targeting JAK1, JAK2, JAK3, and TYK2 to disrupt cytokine-induced activation of the JAK-STAT signaling pathway in the airways. This mechanism reduces pro-inflammatory cytokine and chemokine release in the lungs. Nezulcitinib was primarily investigated for acute lung injury (ALI) associated with COVID-19 in hospitalized patients requiring supplemental oxygen. Clinical trials have shown it to be generally well tolerated with trends toward improved outcomes but without statistically significant efficacy over placebo[1][3][4][5][7].
