Overview

This is a combination therapy of two drugs: - **Paclitaxel** is an anti-mitotic chemotherapy agent that stabilizes microtubules and prevents their depolymerization. This disrupts cell division and induces apoptosis in cancer cells. It is widely used in the treatment of breast cancer and other solid tumors. - **Ruxolitinib** is an orally bioavailable small molecule inhibitor of Janus kinase 1 and 2 (JAK1/2). By inhibiting the JAK-STAT pathway—particularly JAK2-STAT3 signaling—it blocks cytokine-mediated cell proliferation and survival signals. Ruxolitinib has been approved for myelofibrosis, polycythemia vera, and steroid-refractory graft-versus-host disease. The combination has been investigated primarily in HER2-negative metastatic breast cancer—including triple-negative subtypes—and ovarian cancer. Preclinical studies suggest that JAK inhibition can sensitize tumor stem-like cells to chemotherapy by promoting apoptosis through STAT3 pathway inhibition[1][6][8]. Clinical trials have shown this regimen to be tolerable with evidence of clinical activity[1][2].
