Overview
Peficitinib is an orally active small molecule inhibitor of Janus kinases (JAKs), with activity against JAK1, JAK2, JAK3, and Tyk2. It was developed primarily for the treatment of rheumatoid arthritis and has demonstrated efficacy in reducing symptoms and suppressing joint destruction in patients with inadequate response to methotrexate. Peficitinib inhibits the JAK/STAT signaling pathway, thereby modulating immune responses involved in inflammatory diseases. The drug was first approved in Japan in 2019 for rheumatoid arthritis. It has also been investigated for other indications such as psoriasis, ulcerative colitis, and colitis[1][3][5][7][8].
