Overview

PF-06263276 is a small molecule, pan-Janus kinase (JAK) inhibitor developed by Pfizer for inhaled and topical delivery. It was designed using structure-based computational methods to bind beyond the ATP binding site of JAKs, resulting in a type 1.5 binding mode. The drug inhibits multiple JAK isoforms—JAK1, JAK2, JAK3, and TYK2—and was optimized for potency and duration of action suitable for treating inflammatory diseases of the lungs and skin. Clinical development included Phase 1 trials in plaque psoriasis and psoriasis vulgaris.
