Overview

PM116 (also referred to as H116) is a preclinical-stage hydroxyl dendrimer therapeutic (HDT), or 'dendranib,' consisting of a proprietary Janus kinase (JAK) inhibitor conjugated to a hydroxyl dendrimer (HD). Developed by Programmable Medicine and Ashvattha Therapeutics, PM116 is designed to selectively target activated myeloid cells (such as macrophages, dendritic cells, and neutrophils) in inflamed tissues, minimizing systemic exposure and off-target toxicities. In preclinical models of rheumatoid arthritis, PM116 demonstrated significant efficacy in reducing clinical arthritis scores, cartilage damage, pannus formation, inflammation, and bone resorption, comparable to the JAK inhibitor baricitinib.
