POx-Man nanovaccine is a preclinical, mannose-decorated polyoxazoline-poly(lactic-co-glycolic acid) nanoparticle cancer vaccine developed by researchers at Universidade de Lisboa and Tel Aviv University. The approximately 200 nm formulation co-delivers tumor peptide antigens, immune adjuvants, and regulators of transforming growth factor beta expression. Mannose surface functionalization is intended to target the mannose receptor on dendritic cells, improving antigen delivery and induction of antigen-specific, CD8-positive T-cell-mediated antitumor immunity. In mouse colorectal cancer and melanoma models, POx-Man nanovaccines inhibited tumor growth and enhanced the activity of pexidartinib plus PD-1 blockade; these co-administered agents are not intrinsic components of the POx-Man nanovaccine.
