reRNA-25616 is a therapeutic self-amplifying RNA (saRNA) vaccine candidate developed by RNACure Biopharma for the treatment of HPV16-positive solid tumors, including cervical, anal, and head and neck cancers. Utilizing the proprietary reRNA® platform, the vaccine delivers genetic instructions for the expression of HPV16-specific oncoproteins, typically E6 and E7, which are essential for the maintenance of the malignant phenotype in HPV-driven cancers. By inducing host cells to produce these viral antigens, reRNA-25616 stimulates a robust T-cell mediated immune response designed to recognize and eliminate tumor cells. It is currently being evaluated in Phase 1 clinical trials, often in combination with the PD-1 inhibitor toripalimab and the HPV18-targeted vaccine reRNA-22518, to assess its safety, immunogenicity, and preliminary antitumor activity in patients with advanced or metastatic disease.
