Rucotinib phosphate is a Janus kinase (JAK) inhibitor that selectively targets JAK1 and JAK2. It is a phosphate salt formulation of the small molecule rucotinib, which is a phonetic variant of the international nonproprietary name (INN) ruxolitinib. Primarily developed as a topical cream, rucotinib phosphate is utilized for the treatment of dermatological conditions, most notably non-segmental vitiligo. By inhibiting the JAK-STAT signaling pathway, the drug reduces the activity of pro-inflammatory cytokines such as interferon-gamma (IFN-γ), which are implicated in the autoimmune-mediated destruction of melanocytes. Clinical studies, including real-world cohort evaluations in China, have focused on its effectiveness in promoting repigmentation and improving psychosocial outcomes in both adolescents and adults with facial vitiligo.
