Overview
Ruxolitinib + steroids is an investigational combination regimen being evaluated as a first-line therapy for patients with grade II acute graft-versus-host disease (aGVHD) following allogeneic hematopoietic stem cell transplantation. The regimen combines ruxolitinib, a small molecule inhibitor of Janus kinase 1 (JAK1) and Janus kinase 2 (JAK2), with low-dose corticosteroids (typically methylprednisolone). This approach aims to improve response rates compared to steroid monotherapy while minimizing steroid-related toxicities and preventing progression to more severe (grade III-IV) aGVHD. Clinical trials, including phase III studies led by researchers at the Chinese PLA General Hospital, have demonstrated that this combination can achieve high complete response rates and potentially reduce the cumulative dose of steroids required for treatment.
