Overview

Ruxolitinib + chemotherapy is an investigational combination regimen involving ruxolitinib, a selective oral inhibitor of Janus kinase 1 (JAK1) and Janus kinase 2 (JAK2), used together with conventional cytotoxic chemotherapy agents. Ruxolitinib acts by inhibiting the JAK/STAT signaling pathway, which is implicated in tumor cell proliferation, survival, and chemotherapy resistance. Chemotherapy agents commonly used in combinations include paclitaxel, cisplatin, carboplatin, doxorubicin, cyclophosphamide, and topotecan. This combination is being explored primarily for the treatment of solid tumors (notably ovarian and triple-negative inflammatory breast cancer) and hematologic malignancies, aiming to potentiate cytotoxicity and overcome chemoresistance by dual inhibition of signaling pathways and promoting apoptosis[2][4].
