Ruxolitinib + corticosteroids is a combination therapy used primarily in the management of graft-versus-host disease (GVHD), particularly acute GVHD (aGVHD) and chronic GVHD (cGVHD). Ruxolitinib is a selective Janus kinase (JAK) inhibitor, targeting JAK1 and JAK2, which are key mediators in cytokine signaling pathways involved in immune cell activation and proliferation. By inhibiting these kinases, ruxolitinib suppresses pro-inflammatory cytokine production and T-cell expansion. Corticosteroids are broad-spectrum immunosuppressive agents that modulate gene expression to reduce inflammation by binding to glucocorticoid receptors. The combination has shown high response rates as first-line or adjunct therapy for steroid-refractory or high-risk GVHD, with evidence suggesting improved outcomes compared to steroids alone[1][2][3].
