Overview

A combination of ruxolitinib (a small molecule Janus kinase 1 and 2 inhibitor) and crizanlizumab (a monoclonal antibody targeting P-selectin), evaluated in clinical trials for patients with myelofibrosis (MF) who exhibit suboptimal response to ruxolitinib monotherapy. Ruxolitinib disrupts cytokine signaling by inhibiting JAK1 and JAK2, thereby reducing inflammation and hematopoietic proliferation. Crizanlizumab blocks P-selectin on endothelial cells and platelets, inhibiting cell adhesion and potentially decreasing vascular occlusion and inflammation. This combination aims to target both myelofibrosis pathobiology and vascular/inflammatory complications[1][3][5][6].
