Overview

Ruxolitinib + gemcitabine is an investigational combination therapy comprising ruxolitinib, a potent oral inhibitor of Janus kinase 1 and 2 (JAK1/2), and gemcitabine, a nucleoside analog that inhibits DNA synthesis. This combination has primarily been studied in patients with advanced or metastatic solid tumors—including pancreatic cancer and cholangiocarcinoma—where it aims to exploit ruxolitinib’s ability to inhibit oncogenic JAK-STAT signaling, potentially overcoming chemoresistance mediated by pathways like STAT1/STAT3-ALDH1A3 and exerting synergistic antitumor effects with gemcitabine. Ruxolitinib is developed by Incyte and approved as monotherapy for myeloproliferative neoplasms, while gemcitabine is a widely used chemotherapeutic agent. Together, this combination is being evaluated for its safety, tolerability, pharmacokinetics, and preliminary efficacy, with evidence of enhanced cytotoxicity and decreased tumor viability and cell migration in preclinical and early clinical settings[1][2][4].
