Ruxolitinib + gemcitabine + nab-paclitaxel is a combination regimen investigated for the treatment of advanced or metastatic pancreatic cancer and other solid tumors. - **Ruxolitinib** is a small molecule inhibitor targeting Janus kinase 1 and Janus kinase 2 (JAK1/2), which blocks the JAK/STAT pathway implicated in tumor cell proliferation and inflammatory cytokine production. - **Gemcitabine** is a nucleoside analog that inhibits DNA synthesis, resulting in cytotoxicity against rapidly dividing cells. - **Nab-paclitaxel** is a nanoparticle albumin-bound formulation of paclitaxel, which stabilizes microtubules and disrupts mitosis in tumor cells. Nab-paclitaxel increases intratumoral gemcitabine levels, potentially enhancing efficacy. This combination was studied in a Phase Ib trial for advanced solid tumors, primarily pancreatic cancer. The regimen showed a tolerable safety profile with reduced doses but did not reach full clinical development as the sponsor terminated the study after interim analysis due to insufficient efficacy observed in related trials[1][3][5].
