Overview

DEP-Ru is a **co-administration salvage regimen** for refractory or relapsed hemophagocytic lymphohistiocytosis comprising oral ruxolitinib plus liposomal doxorubicin, etoposide, and methylprednisolone. Ruxolitinib inhibits JAK1 and JAK2 to suppress cytokine-driven hyperinflammation, while liposomal doxorubicin and etoposide provide cytotoxic and topoisomerase II-directed activity and methylprednisolone provides glucocorticoid-mediated immunosuppression. Beijing Friendship Hospital, Capital Medical University evaluated the regimen in a multicenter, single-arm phase 2 trial for refractory or relapsed hemophagocytic lymphohistiocytosis.
