Overview

Ruxolitinib is a potent and selective small molecule inhibitor of Janus kinase 1 (JAK1) and Janus kinase 2 (JAK2), developed for the treatment of myeloproliferative neoplasms such as myelofibrosis, polycythemia vera, and graft-versus-host disease. It works by blocking dysregulated JAK signaling involved in abnormal blood cell proliferation and inflammation[3][4][5][6]. Simvastatin is an oral HMG-CoA reductase inhibitor used to lower cholesterol levels, reduce cardiovascular risk, and has additional anti-inflammatory properties[2]. The combination of ruxolitinib plus simvastatin has been investigated for synergistic effects in conditions characterized by excessive inflammation, such as COVID-19, where both drugs may help prevent respiratory deterioration through immunomodulation and anti-inflammatory actions[1].
