RVU120 + ruxolitinib is an investigational combination therapy comprising RVU120, a highly selective and potent cyclin-dependent kinase 8 and 19 (CDK8/19) inhibitor, and ruxolitinib, a Janus kinase 1 and 2 (JAK1/2) inhibitor. The combination is currently being studied for treating myelofibrosis (MF), with both preclinical and early clinical evidence indicating biochemical and functional synergy. RVU120 acts by selectively inhibiting CDK8/19, thereby modulating STAT phosphorylation independently of JAK1/2, while ruxolitinib inhibits aberrant JAK-STAT signaling driving disease progression in MF and other myeloproliferative neoplasms (MPNs). The combination shows promise through dual targeting of the JAK-STAT pathway, particularly in MF patients with suboptimal response to JAK inhibition, and is under Phase II clinical investigation.
