SOCS1-KIR is a synthetic peptide mimetic of the kinase inhibitory region (KIR) of the Suppressor of Cytokine Signaling-1 (SOCS1) protein. It functions as an intracellular negative regulator of the JAK/STAT signaling pathway, specifically inhibiting signaling induced by interferon-gamma (IFN-γ). The peptide works by binding to the activation loop of Janus kinases (JAKs), preventing the subsequent phosphorylation and activation of STAT transcription factors. Developed by researchers at the University of Florida, SOCS1-KIR has shown potential in preclinical models of systemic lupus erythematosus (SLE) to reduce autoantibody production, kidney pathology, and skin lesions while promoting a more regulatory immune environment by enhancing Foxp3 expression in regulatory T cells.
